چکیده

Haemagglutinin and neuraminidase have clearly been shown to be responsible for the stimulation of immunity against influenza. In the approach described, these two immunogens are selectively solubilized, leaving an intact subviral particle which contains lipid and non-protective components. The methods used can readily be adapted to large scale production. The immunogenicity of such antigen-preparation was studied in different animal species. Antibodies against haemagglutinin and neuraminidase were measured in mice, hamsters, guinea-pigs and rabbits. In addition, mice-protection experiments were performed. The immune response of volunteers to this type of subunit vaccine has also been evaluated (Kunz and Hofmann, 1975). Results of a first field trial indicate that the solubilization procedure did not alter the immunogenic properties of the two antigens.

Cloning, expression and purification of hemagglutinin conserved domain (HA2) of influenza A virus, to be used in broad-spectrum subunit vaccine cocktails

Introduction: Influenza virus has several conserved peptides which have the capacity to be used as suitable candidates for appropriate and stable vaccine production against different types of influenza viruses. One of these peptides is HA2, the hemagglutinin stalk domain which mediates the membrane fusion and is conserved amongst different sub-types of influenza virus. This peptide is a good ca...

Protective Immunity in Mice Following Immunization with the Cochleate-Based Subunit Influenza Vaccines

High morbidity and mortality of influenza virus infection makes it an important disease world-wide. Mouse is a very well-studied animal model for this disease with similar manifestation to human disease. It would be desirable to induce mucosal as well as circulating immune responses to obtain protection from infection and to decrease the spread of the virus. Cell mediated immunity (proliferativ...

Subunit Vaccine Preparation of Bovine Rotavirus and Its Efficacy in Mice

Background: Rotaviruses (RV) are important viral diarrheal agents in calves. Vaccination is an optimum measure to prevent bovine rotaviruses (BRV) infection. However, little research on BRV VP7 vaccine has been done and currently there is no BRV vaccine. Objective: To prepare a subunit vaccine of BRV and investigate its efficacy. Methods: Total RNA was extracted from MA104 cells infected with b...

Challenges and Perspectives toward Development of more Effective Influenza Vaccine

Influenza viruses continue to be a major health threat in human and bird populations. The improvements in formulation and production level of the current influenza vaccines are not sufficient to afford complete protection. The continuous antigenic drifts and emergence of endemic and zoonotic strains make influenza vaccine planning difficult. Concern about the emergence of new influenza pandemic...
